Semen samples were collected from 2 elephant bulls (EM 1 and EM 2;4 ejaculates per bull) by manual manipulation. Semen were diluted with TEST containing 5% glycerol final concentration frozen in liquid nitrogen after 0,1,2,3,4 and 5 hours of equilibration times. The frozen semen were thawed and evaluated for progressive motility, viability and acrosome integrity. In EM 1, the percentage of progressive motility of frozen-thawed spermatozoa after equilibrated for 0,1,2 and 3 hours did not differ from those found after equilibration for 4 and 5 hours. In EM2, the percentage of progressive motility of frozen-thawed spermatozoa after 0,1,2 and 3 hours. In EM 2, the percentage of progressive motility of frozen-thawed spermatozoa after 0,1,2 and 3 hour equilibrations were significantly higher (p<0.05) than those after 4 and 5 hour eqilibrations. The different equilibration time (o-5h) did not effect on viability and acrosome integrity of frozen-thawed spermatozoa. This finding indicates that the equilibration times prior freezing affected to frozen-thawed sperm motility with the trend that higher motie spermatozoa were obtained after 0-3 hour equilibrations. The obtained knowledge will be aided in the development of cryopreservation technique and artificial insemination in Asian elephants in the future. |
